How it does it. Semax is a peptide of seven amino acids. It takes a short fragment of ACTH, one of the body's own hormones, and adds a three amino acid tail that stops enzymes breaking it apart. Semax (CAS 80714-61-0; molecular formula C37H51N9O10S, average molecular weight ≈ 814 g/mol; sequence MEHFPGP) is a synthetic heptapeptide derived from the ACTH(4-10) fragment, extended at the C-terminus with a Pro-Gly-Pro motif that stabilises the peptide against enzymatic degradation. It is referenced as the leading pro-cognitive nootropic research peptide in the literature.

Its neurotrophic mechanism has been characterised in research models: Dolotov and colleagues (Brain Research, 2006) reported that Semax raises hippocampal BDNF (brain-derived neurotrophic factor) protein and exon-specific BDNF mRNA and increases trkB-receptor phosphorylation, implicating the BDNF/trkB signalling system.

A companion study by Dolotov and colleagues (Journal of Neurochemistry, 2006) demonstrated specific, saturable binding of the peptide together with a rapid rise in basal-forebrain BDNF protein — evidence for a receptor-mediated neurotrophic mechanism. These anchor Semax as a reference peptide for BDNF-pathway research.
